苹果砧木‘青砧一号’离体高效再生体系的建立

摘 要:

‘青砧一号’是杂交育成的苹果矮化砧木新品系。本文探讨了影响‘青砧一号’离体再生的条件, 建立了高效离体再生体系。结果表明, 以叶片为外植体, 远轴面接触培养基, 最佳不定芽诱导分化培养基: MS+5.0 mg·L-1 6-BA+0.3 mg·L-1 NAA; 最佳继代增殖培养基: MS+1.0 mg·L-1 6-BA+0.1 mg·L-1 IAA+20 g·L-1蔗糖, 不定芽的增殖倍数为2.7; 最佳生根培养基: 1/2MS+1.0 mg·L-1 IAA+20 g·L-1蔗糖, 生根率达100%, 平均生根数达5.7条。炼苗后移入珍珠岩、蛭石、草炭等体积混合的基质中, 20 d后移栽成活率达60%。

Establishment of Efficient in vitro Regeneration System of Apple Rootstock ‘QZ-1’ (Malus hupehensis×M. domestica)

Abstract:

‘QZ-1’ (Malus hupehensis×M. domestica) is a new strain of apple dwarfing rootstocks created by hybridization. In this experiment, the factors influencing leaf regeneration of ‘QZ-1’ were studied and the efficient in vitro regeneration system of ‘QZ-1’ was established. The results showed that MS medium with 0.3 mg·L-1 NAA and 5.0 mg·L-1 6-BA was optimal for adventitious bud differentiation. The best medium for subculture proliferation was MS supplemented with 1.0 mg·L-1 6-BA, 0.1 mg·L-1 IAA and 20 g·L-1 sucrose, and the proliferation times of adventitious buds reached 2.7 on it. 1/2MS medium containing 1.0 mg·L-1 IAA and 20 g·L-1 sucrose was the most suitable for rooting. The rooting rate was up to 100%, and the average number of rooting reached 5.7. After acclimatization, the regenerated plants were transplanted into the matrix, which contained perlite, vermiculite and peat (1:1:1, V:V:V). The survival rate was up to 60% after 20 d.
